I'm fairly certain that I had my mouse in the serial port. Now it won't work there. Why not?

You could check for pin integrity, contact corrosion and whether or not your serial card (if applicable) has been jostled out of the slot somehow.

Apart from that I wouldn't know what to suggest looking for.  Before I obtained an optical mouse I went through 2-3 mice a year.

All it took was a reboot.

well, some computers have 2 serial ports, so check that..but if you are still using a computer that uses a serial mouse, might i suggest a new computer

After a little more fiddling, I was able to get the mouse plugged into the regular mouse port. Damn adapters were in the way. But it *was* in the serial port.

A little knowledge is a dangerous thing. Your problem is that you know which one is the serial port. ;~)
